Background: Non-alcoholic Fatty Liver Disease (NAFLD) is the most common liver disease in
industrialized countries. Defective autophagy of lipid droplets in hepatocytes, also known as
lipophagy, has been recently indicated as a possible pathophysiological mechanism of NAFLD.
Experimental and epidemiological evidence suggest that dietary polyphenols, such as
flavonoids may prevent NAFLD. To address this hypothesis and analyze the underlying
mechanisms we supplemented Bergamot Polyphenol Fraction (BPF) to “cafeteria”-diet-fed rats.
“Cafeteria” diet (CAF) accurately reflects the variety of highly palatable, energy dense foods
consumed by children in industrialized countries. Feeding rats with CAF diet rapidly induces
hepatic steatosis and features of metabolic syndrome and currently, CAF diet is considered the
best animal model for these pathologies.
Methods and Results: BPF treatment (50 mg/kg daily, supplemented with drinking water)
potently counteracted the pathogenic increase of blood triglycerides and had minor effects on
blood glucose and obesity, observed in this animal model. Importantly, BPF showed strong
effect on hepatic steatosis documented by ultrasound examination and histological analysis of
liver sections two and three months post-treatment, respectively. The morphometric analysis
of oil-red stained sections confirmed a mean 48.5% reduction in total area and in numbers of
all categories of lipid droplets accumulated in hepatocytes. BPF-treated liver lysates showed
increased levels of LC3I and Beclin-1 and reduction of p62, suggesting autophagy stimulation
by BPF. Higher levels of LC3I and II in the lipid droplet subcellular fractions, as well as colocalization
of LC3 with Tip47-positive and oil-red stained vesicles in livers from BPF-treated
animals indicated lipophagy stimulation by polyphenols present in BPF. This was confirmed by
in vitro studies. BPF dose-dependently enhanced autophagic flux and AMP kinase activation in
HepG2 cells. This correlated with the ability of BPF to counteract the accumulation of palmitic
acid-induced lipid vesicles. Further experiments suggested that the pro-autophagic activity of
BPF is exerted mainly by aglycon forms of bergamot flavonoids.
Conclusions: This study suggests that the liver and its lipid metabolism are the main targets
of bergamot polyphenols, indicating BPF-diet supplementation as an effective strategy to
prevent NAFLD. Beneficial effects of BPF can be explained by its ability to stimulate hepatic
lipophagy and AMP kinase activation.

Mechanisms Underlying the Anti-Tumoral Effects of Citrus bergamia Juice

Abstract
Based on the growing deal of data concerning the biological activity of flavonoid-rich natural products, the aim of the present study was to explore in vitro the potential anti-tumoral activity of Citrus Bergamia (bergamot) juice (BJ), determining its molecular interaction with cancer cells. Here we show that BJ reduced growth rate of different cancer cell lines, with the maximal growth inhibition observed in neuroblastoma cells (SH-SY5Y) after 72 hs of exposure to 5% BJ. The SH-SY5Y antiproliferative effect elicited by BJ was not due to a cytotoxic action and it did not induce apoptosis. Instead, BJ stimulated the arrest in the G1 phase of cell cycle and determined a modification in cellular morphology, causing a marked increase of detached cells. The inhibition of adhesive capacity on different physiologic substrates and on endothelial cells monolayer were correlated with an impairment of actin filaments, a reduction in the expression of the active form of focal adhesion kinase (FAK) that in turn caused inhibition of cell migration. In parallel, BJ seemed to hinder the association between the neural cell adhesion molecule (NCAM) and FAK. Our data suggest a mechanisms through which BJ can inhibit important molecular pathways \related to cancer-associated aggressive phenotype .

Each individual practitioner has his own beliefs in their products. The process I explained above of FDA approval cannot be denied. While I acknowledged the "trials", at the end of the day, it's still a vitamin. Even daily vitamins have clinical literature supporting their use. Like I said, it's no better than vino lowering one's blood pressure. Its no better than acai berry claim to losing weight. It's the law of the land Tazzy. It'll never get FDA approval. Doesn't need to, it's not a chemically modified substance which claims to cure disease. It's a natural supplement that is only used in combination with other medications. Call me an asshole all you want & you can stutter your way until you pass out, but at the end of the day it would never be a blockbuster drug. However, I can teach you a trick. Find out how Lovaza got approved for prescription use and their manufacturing practices and you may have a shot at something. 1200mg of Fish Oil, branded and sold millions of dollars.
